Biochemists test fluid samples to see if hormones, substances, antibodies proteins, or other antigens are present in the solution. ELISA reader is a specialized spectrophotometer to measure light intensity and allow biochemists to measure the reactions of antigens and antibodies in a solution through enzyme activity. The ELISA is performed in a several well polystyrene plate in one of two formats: either antigen is attached to the bottom of each well, or the capture antibody is attached to the bottom of each well.
A typical microplate contains 8 columns and 12 rows for 96 sample wells. It allows you to check up to 96 different conditions simultaneously. The automated ELISA reader has an optical system that analyzes samples with light so that users can interpret the results.

Indirect ELISA


Indirect ELISA works with a two-step process. First, a primary antibody is added to the sample. A secondary antibody is then added to bind to the primary antibody. This method has high flexibility and signal amplification. However, it may result in cross-reaction from the secondary antibody, making the result inconclusive.

Direct ELISA


During the direct ELISA test, the antigen is added directly to the microplate well. The antibody is then added to the solution and binds directly to the antigen to prepare it for the addition of another enzyme. Finally, the next test enzyme is added and analyzed by the color change of the test. Advantages of this test include fewer steps, fewer reagents needed, and no secondary antibody reactions that can confound the results.
